Mutational Signature-Based Biomarker for Phase II Trial of Olaparib Maintenance in Advanced High-Grade Ovarian Cancer.
This phase 2 trial developed and validated a mutational signature-based biomarker (MSBM) to define homologous recombination deficiency (HRD) for olaparib maintenance in newly diagnosed advanced high-grade ovarian cancer. In 170 patients with stage IIIC/IV disease, MSBM integrated three whole-exome sequencing-derived features (tumor BRCA mutation status, Mutational Signature 3 similarity score >0.5, or score >0.25 without CCNE1 amplification) to identify HRD-positive patients for olaparib maintenance efficacy assessment (58 HRD-positive patients responding [truncated]). If validated, MSBM could enable earlier and more practical HRD selection for olaparib by avoiding genomic scar-based assays.

Tumor-Derived Extracellular Vesicles in Plasma for Predicting Anti-PD-1 Antibody Efficacy in Non-Small Cell Lung Cancer.
This study developed plasma liquid-biopsy antibody-based sandwich assays to quantify extracellular vesicle (EV) subsets expressing cell surface vimentin (CSV) and co-expressing EpCAM or PD-L1, and tested whether baseline EV levels predict anti-PD-1 efficacy in non-small cell lung cancer (NSCLC). In 57 resected stage I NSCLC patients and 89 stage IV NSCLC patients treated with anti-PD-1 antibodies, baseline EpCAM/CSV-EV levels and related EV subset measurements were associated with clinicopathological features and treatment response. These findings support CSV/EpCAM- and CSV/PD-L1–positive EVs as potential predictive biomarkers for selecting patients likely to benefit from anti-PD-1 therapy.

Diagnostic performance of droplet digital PCR for KRAS mutation analysis in pancreatic cancer cytology specimens.
This study evaluated droplet digital PCR (ddPCR) for KRAS mutation detection in residual cytology wash fluid from endoscopic ultrasound–guided fine-needle aspiration (EUS-FNA) in 38 patients with pancreatic cancer, benchmarking ddPCR against next-generation sequencing (NGS) of tumor tissue. ddPCR detected KRAS mutations with a lower positivity threshold (0.1%) than tissue NGS (1.0%) and showed concordance between cytology-based ddPCR and tissue-based NGS. Rapid, tissue-sparing KRAS genotyping from cytology wash fluid could enable timely selection of patients for KRAS-targeted therapies when tissue is limited.

An HDAC4-specific PROTAC degrader achieves radiation sensitization by enhancing ferroptosis in lung cancer.
This study examined how HDAC4 regulates ferroptosis and radiotherapy response in lung cancer, using CRISPR screening in patient-derived organoids and mechanistic assays of HDAC4-mediated post-translational modification. HDAC4 promoted radiation resistance by acting as an E3 SUMO ligase that SUMOylated MBD1, preventing MBD1 ubiquitination/degradation and thereby repressing TP53 and CYP1A1 transcription to suppress lipid ROS and ferroptosis; an HDAC4-specific PROTAC (TP1) enhanced radiation sensitization by restoring ferroptosis. These findings identify an HDAC4–MBD1–TP53/CYP1A1 axis as a therapeutic vulnerability and support PROTAC-mediated HDAC4 degradation as a strategy to improve lung cancer radiotherapy outcomes.

Efficacy of Cabozantinib in Patients With Advanced Renal Cell Carcinoma With Bone Metastasis Assessed by Modified MD Anderson Criteria: An Open-Label Phase 2 Study.
This open-label phase 2 study evaluated cabozantinib efficacy in adult Japanese patients with advanced renal cell carcinoma (aRCC) with bone metastasis, using a modified MD Anderson criteria to assess objective response rate (ORR) of bone target lesions. Cabozantinib produced measurable responses and disease control in bone lesions, with secondary outcomes including overall survival (OS) and progression-free survival (PFS). A positive feedback loop between BACH1 and IL-1β promotes the progression of HPV-negative head and neck squamous cell carcinoma.
This study investigated the role of BTB Domain and CNC Homolog 1 (BACH1) in HPV-negative head and neck squamous cell carcinoma (HNSCC) progression. The key finding was that BACH1 is upregulated in HPV-negative HNSCC and promotes tumor progression by enhancing proliferation and inhibiting apoptosis, in part by associating with the SWI/SNF chromatin remodeling complex and transcriptionally activating IL1B. Macrophage exosome-modified Mn/Se nanoheterostructure induces post-infarction cardiomyocyte regeneration and immunometabolic microenvironment repair.

Real-life experience in the treatment of gallbladder carcinoma by transarterial radioembolization: a multicenter retrospective study.
This multicenter retrospective study evaluated transarterial radioembolization for gallbladder carcinoma with liver-dominant metastatic disease, using yttrium-90 or holmium-166 radioembolization across five tertiary centers. Single-nucleus profiling of postmortem diffuse midline gliomas identifies mitochondrial biogenesis as a resistance mechanism to imipridone therapy.
This study used single-nucleus RNA sequencing and open-chromatin sequencing on postmortem tissue from 22 diffuse midline glioma (DMG) cases treated with imipridones (including ONC201/imipridone) to identify resistance mechanisms, with additional myeloid phenotyping in 46 cases. The key finding was that mitochondrial biogenesis emerged as a tumor-intrinsic resistance mechanism to imipridone therapy. Fructose 1-phosphate inhibits mannose phosphate isomerase to suppress hepatocellular carcinogenesis.
The study examined how fructose suppresses hepatocellular carcinoma (HCC) in the setting of fructolytic enzyme loss, focusing on fructose-1-phosphate (F1P) inhibition of mannose phosphate isomerase (MPI) in ALDOB-deficient contexts. It found that fructose-derived F1P inhibits MPI to suppress hepatocellular carcinogenesis when ALDOB deficiency is present, supported by transcriptomic and metabolomic analyses (metabo[…]). Multifunctional spatiotemporally programmed microneedle patches for the reconstruction of antibacterial and immunoregenerative homeostasis in periodontitis therapy.

Efficacy of Upadacitinib in Patients with Atopic Dermatitis of the Head and Neck Region.
This post hoc analysis of Measure Up 1 (NCT03569293) and Measure Up 2 (NCT03607422) studied efficacy of upadacitinib versus placebo in patients with head and neck atopic dermatitis, including stratification by baseline head and neck EASI. At week 16, upadacitinib (15 mg and 30 mg) produced higher rates of stringent clearance targets (e.g., head/neck EASI ≤0.3, EASI 90/100) and improved patient-reported outcomes compared with placebo (full results truncated). The findings support upadacitinib as an effective targeted therapy for difficult-to-treat head and neck AD.

Real-world efficacy and safety of immune checkpoint blockades in advanced urothelial carcinoma with histologic variant subtypes: the pembroblad study.
This French multicenter retrospective study (pembroblad) examined real-world outcomes of immune checkpoint blockade in advanced urothelial carcinoma with histologic variant subtypes (UC-V) and non-urothelial carcinomas (NUC) after platinum progression. Patients received second-line anti–PD-(L)1 monotherapy between 2016 and 2022, and the study evaluated overall response and safety across these under-represented histologic variant groups (primary endpoint details truncated). The results are clinically important for guiding ICB use in aggressive bladder/urinary tract cancers that are poorly represented in randomized trials.

PSMC2 promotes hepatocellular carcinoma progression through interaction with EGFR.
This study examined the role of Proteasome 26S Subunit ATPase 2 (PSMC2) in hepatocellular carcinoma (HCC) progression and its interaction with EGFR using TCGA analyses, immunohistochemistry (IHC), Western blot, and HCC cell functional assays. PSMC2 was upregulated in HCC and promoted malignant phenotypes (proliferation, colony formation, migration, and invasion) while mechanistically interacting with EGFR to drive tumor progression. These findings suggest that the PSMC2–EGFR axis could be a therapeutic target or biomarker pathway for HCC.

Microglial MARCO facilitates Varicella zoster virus uptake and triggers TLR2-mediated neuroinflammation.
This study examined how varicella zoster virus (VZV) affects microglial morphodynamics and function in human microglial models (HMC3, HIM, and human embryonic stem cell–derived microglia) using wild-type VZV (YC01) and attenuated strains (MAV/06, MAV). VZV uptake was facilitated by microglial MARCO and triggered TLR2-mediated neuroinflammation, supported by bulk RNA sequencing. These findings identify the MARCO–TLR2 axis as a mechanistic driver of VZV-induced neuroinflammation and a potential therapeutic target to limit CNS damage during infection.

Comparative fall risk of patients treated with novel androgen receptor antagonists in prostate cancer: a systematic review and meta-analysis.
This systematic review and meta-analysis evaluated fall risk in patients with prostate cancer treated with novel androgen receptor pathway inhibitors (enzalutamide, apalutamide, darolutamide) versus placebo or non-steroidal antiandrogens in Phase 2/3 randomized controlled trials. Across trials, the key outcome was risk ratios for all-grade and grade ≥3 falls, enabling drug-specific comparison of ARPI-associated fall risk. Clinically, the results support more informed selection and monitoring of ARPIs by quantifying which agents may carry higher fall risk in advanced prostate cancer.

Pulmonary toxocariasis presenting as migratory pulmonary infiltrates and mediastinal lymphadenopathy: a case report and literature review.
This case report and literature review described pulmonary toxocariasis in a 53-year-old female smoker presenting with migratory pulmonary infiltrates and mediastinal lymphadenopathy that mimicked lung cancer. Despite non-diagnostic bronchoscopy with EBUS-TBNA, the patient was treated empirically with dexamethasone for suspected organizing pneumonia, reflecting the diagnostic challenge of eosinophilic lung disease caused by Toxocara canis/cati. The report highlights the need to consider toxocariasis in the differential diagnosis of eosinophilic pulmonary presentations with malignancy-like imaging.

Association between preoperative quality of life (QoL) and postoperative complications after gastrectomy for gastric carcinoma.
This retrospective analysis assessed whether preoperative quality of life (QoL) predicts postoperative complications after gastrectomy for gastric carcinoma in 908 prospectively collected patients. Preoperative QoL was measured using EORTC QLQ-C30 and STO22, and multivariate logistic regression with ROC evaluation was used to build a predictive model. If validated, the work supports using baseline QoL instruments to stratify surgical risk and anticipate complication likelihood after gastrectomy.

Osteopontin-driven TGF-β1 signaling via integrin αvβ3-NF-κB axis impairs NK cell function in head and neck squamous cell carcinoma.
The study investigated how osteopontin (OPN)-driven TGF-β1 signaling through the integrin αvβ3–NF-κB axis impairs natural killer (NK) cell function in head and neck squamous cell carcinoma (HNSCC). Using flow cytometry, multiplex immunohistochemistry, and RNA-seq analyses (bulk and single-cell), it found that OPN–integrin αvβ3/NF-κB signaling promotes TGF-β1 production that suppresses NK cell activity. Obesity suppresses neutrophil-dependent itaconate signaling promoting ferroptosis in acute pancreatitis.
This mechanistic study investigated how obesity alters neutrophil-dependent itaconate signaling to promote ferroptosis in acute pancreatitis, focusing on myeloid-derived itaconate delivery to pancreatic acinar cells. Obesity enhanced a pathway in which neutrophil/myeloid-derived itaconate, transferred via the SLC13A3 transporter, modulated NRF2-dependent antioxidant responses that protect against ferroptosis during pancreatic inflammation (with obesity disrupting protective regulation; details truncated). ZNF831 suppresses triple-negative breast cancer progression through NLRP3-associated pyroptotic signaling and M1-like macrophage phenotypic remodeling.
This study investigated whether zinc finger protein 831 (ZNF831) suppresses triple-negative breast cancer (TNBC) progression by modulating NLR family pyrin domain containing 3 (NLRP3)-associated pyroptotic signaling and remodeling macrophage phenotypes, using integrated TCGA-BRCA and Gene Expression Omnibus bulk datasets plus mechanistic experiments. The key finding was that ZNF831 enhances NLRP3-linked pyroptosis and drives M1-like macrophage phenotypic remodeling, correlating with improved antitumor immune activity in TNBC. Vaccine-expanded plasmablast-like B cells are associated with response to dendritic cell therapy in metastatic melanoma.
This study analyzed vaccine-expanded plasmablast-like B cells within dendritic cell vaccine (DCVax) products and related immune features to determine associations with clinical response in patients with metastatic melanoma receiving monocyte-derived DC therapy. The key finding was that higher frequencies/abundance of vaccine-expanded plasmablast-like B cells were associated with responder status compared with non-responders. Spatial and single-cell transcriptomic profiling reveals interplay between SRGN-associated epithelial programs and C1QC+ macrophages in early LUAD progression.
This study used integrated single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics to map early lung adenocarcinoma (LUAD) progression and specifically examined the interplay between SRGN-associated epithelial programs and C1QC+ macrophages across stages from precursor lesions to invasive disease. The key finding was that SRGN-linked epithelial states co-evolve with C1QC+ macrophage programs, indicating coordinated epithelial–macrophage crosstalk during early LUAD progression. Mast cell-specific receptor MrgprB2 selectively mediates oxaliplatin-induced neuropathic pain.
This study investigated in mice how mast cell-specific MrgprB2 signaling mediates oxaliplatin-induced chemotherapy-induced peripheral neuropathic pain, comparing oxaliplatin versus paclitaxel effects and focusing on the mast-cell receptor MrgprB2 (human orthologue MrgprX2). Lactic acid induces dendritic cell pyroptosis through MCT-1 to promote tumor immune evasion.
This study investigated how lactic acid in the tumor microenvironment affects dendritic cell (DC) survival and function, using bone marrow-derived DCs (BMDCs) to model DC pyroptosis. Lactic acid induced dose-dependent pyroptosis via GSDMD cleavage through monocarboxylate transporter 1 (MCT1)-mediated signaling along the K+/NLRP3/GSDMD axis, promoting tumor immune evasion. Targeting MCT1 or the NLRP3/GSDMD pyroptotic pathway could restore DC-mediated anti-tumor immunity in lactic-acid–rich tumors.

RFX5 as a regulator of spatial chromatin contacts and gene expression.
This study identified RFX5 as a regulator of spatial chromatin contacts and gene expression by performing CRISPR-mediated RFX5 knockout in A375 cancer cells followed by RNA-seq, ChIP-seq, ATAC-seq, QHR-4C, and Hi-C. RFX5 loss reduced chromatin accessibility and loop strength, decreased CTCF/RAD21 occupancy, and suppressed expression of ~2,000 genes including cancer-associated oncogenic pathways. RFX5 therefore emerges as a CTCF/RAD21-associated factor that helps maintain 3D genome organization and oncogenic transcriptional programs.

SCRUM-Japan MONSTAR3 hematology cohort: a nationwide multi-omics integrated platform for next-generation precision medicine in hematologic malignancies.
SCRUM-Japan MONSTAR3 established a nationwide, prospective multi-omics integrated platform (SCRUM-Japan MONSTAR3 hematology cohort) enrolling patients with newly diagnosed or relapsed/refractory hematologic malignancies. The cohort collects tumor specimens (bone marrow aspirates/biopsies or lymph node tissues) at diagnosis and at subsequent timepoints to enable integrated genomic and other omics analyses for precision medicine. Signals from the bone marrow B cell niches shape pre-leukemic fate in a murine model of B cell acute lymphoblastic leukemia.
This work studied how bone marrow (BM) B cell niche signals shape pre-leukemic fate in a murine model of B cell acute lymphoblastic leukemia (B-ALL), focusing on pre-B cell receptor (pre-BCR) signaling. The microenvironmental blockade of the pre-BCR ligand Galectin-1 impaired pre-leukemic cell proliferation and leukemia-initiating capacity, indicating niche control over pre-leukemic fitness. These findings highlight BM niche–dependent mechanisms that could be exploited to prevent progression from pre-leukemic states to overt B-ALL.

Human retinal organoids functionally bridge a transected optic nerve in a rat model.
This study tested whether heterotopic transplantation of human retinal organoids into the transected optic nerve cavity can restore vision in adult rats. One month after transplantation, treated rats showed partial, sustained functional recovery (direct pupillary light reflex, visually evoked electrophysiology, and visually guided behaviors for at least 3 months) alongside neuroanatomical evidence of structured host–graft coupling across the gap. The work provides a translationally relevant bridge between retinal tissue and a severed optic nerve, supporting organoid-based circuit reconstruction for adult mammalian vision restoration.

Neocortical tau burden determines the degree of cognitive impairment in individuals with Braak stage V neurofibrillary degeneration.
This study examined whether neocortical tau burden predicts the degree of cognitive impairment in humans with Alzheimer disease neuropathologic change at Braak stage V. Using quantitative computer-assisted positive-pixel assessments to measure neocortical hyperphosphorylated tau (p-tau) burden in individuals with Braak V, the authors found that neocortical tau burden better explained variability in cognitive impairment than existing neuropathologic metrics alone (details truncated in the provided abstract). Lipoprotein(a) Levels, Risk of Cardiovascular Events and Benefit of Evolocumab: Findings From the VESALIUS-CV Trial.
This controlled study assessed the prevalence and clinical correlates of Rome IV-defined functional bowel disorders (FBDs) in idiopathic Parkinson’s disease (iPD). CHCHD2 and CHCHD10 promoted autophagic clearance of protein aggregates via GABARAPs.

Protein Disulfide Isomerase Disassembles TDP-43/G3BP1 Condensates and Antagonizes TDP-43 Pathological Aggregates.
The study examined how protein disulfide isomerase (PDI) modulates TDP-43 pathology by disassembling TDP-43/G3BP1 condensates and antagonizing pathological TDP-43 aggregates. Wild-type PDI interacted with TDP-43 to attenuate TDP-43 phase separation, competitively displaced G3BP1 to disassemble condensates, and counteracted pathological mislocalization and abnormal phosphorylation. These mechanistic findings suggest PDI as a potential therapeutic target to reduce TDP-43 aggregation relevant to ALS and TDP-43 proteinopathies.

NMN/NAD+ enhances SIRT2-modulated microtubule dynamics to improve mitochondrial and mitophagy functions in senescent cells.
This study tested whether NMN (nicotinamide mononucleotide)/NAD+ enhances SIRT2-mediated microtubule dynamics to improve mitochondrial function and mitophagy in senescent human cells. NMN/NAD+ increased SIRT2-dependent regulation of microtubule stability/dynamics, leading to improved mitochondrial performance and enhanced mitophagy in senescent cells. Scientifically, it supports an NAD+-SIRT2-microtubule axis as a targetable pathway to counter senescence-associated mitochondrial dysfunction.

Neural mesenchymal-like fibroblast-derived migrasomes promote Schwann cell proliferation and migration via tenascin-C transfer during peripheral nerve regeneration.
This study examined how neural mesenchymal-like fibroblast-derived migrasomes influence Schwann cell (SC) behavior during peripheral nerve regeneration. The key finding was that after peripheral nerve injury, neural mesenchymal-like fibroblasts expand and produce migrasomes that promote SC proliferation and migration and provide directional cues, with tenascin-C transfer implicated as the mechanism. Assessing the role of Subacute Encephalopathy with Seizures in Alcoholics (SESA) syndrome in the development of status epilepticus: a case report and systematic review with individual patients’ analysis.
This article reported a super-refractory status epilepticus (SRSE) case in a patient with confirmed Subacute Encephalopathy with Seizures in Alcoholics (SESA) and also performed a systematic review of SESA progression to status epilepticus. The key finding was that SESA can progress to SRSE, but the relationship between SESA and status epilepticus remains incompletely characterized in the literature. Clinically, it highlights the need for heightened monitoring and early aggressive management when SESA patients deteriorate toward refractory status epilepticus.

ERVK activity in CD8+ T cell immune cell compartment in patients with ALS.
This study examined endogenous retrovirus-K (ERVK) integrase (IN) expression in immune-cell compartments by analyzing peripheral blood mononuclear cells (PBMCs) from patients with amyotrophic lateral sclerosis (ALS), focusing on lymphoid and myeloid-derived immune cells. The key finding was that ERVK IN is expressed in these immune cells, supporting a link between ERVK activity and ALS-associated genomic instability in the immune system. In vivo profiling of astrocyte secretome reveals brain-region specific regulatory networks in a mouse model of amyloid pathology.
This study used in vivo TurboID proximity labeling to profile the astrocyte secretome in a mouse model of amyloid pathology, mapping brain-region specific regulatory networks across disease progression. Engineered neuronal exosomes mediate α-synuclein clearance to ameliorate Parkinson’s disease.
This study developed engineered neuronal exosomes loaded with an α-synuclein (αSyn) aggregation-blocking peptide (sPep) to enhance αSyn clearance and test therapeutic effects in a Parkinson’s disease (PD) context. The key finding was that engineered exosomes extended blood circulation half-life to 3.8 hours, improved brain targeting (2.15% brain signal vs 0.78% for free dye), and were designed to block αSyn aggregation. This supports engineered exosome delivery of aggregation-inhibiting peptides as a promising disease-modifying strategy for PD.

Deficient TRPM3-linked mitochondrial Ca2+ influx in natural killer cells associated with myalgic encephalomyelitis/chronic fatigue syndrome.
This study investigated ME/CFS pathomechanisms by examining TRPM3-linked mitochondrial Ca2+ influx in natural killer (NK) cells from individuals with ME/CFS, building on prior evidence of impaired TRPM3-dependent Ca2+ mobilization. Using fluorescence live-cell imaging, the authors assessed downstream mitochondrial Ca2+ handling resulting from TRPM3 dysfunction in NK cells. Proximity proteomics reveals a co-evolved LRRK2-regulatory network linked to centrosomes.
This study used BioID proximity proteomics to map LRRK2-associated protein networks and applied evolutionary/structural bioinformatics to interpret co-evolved interactions in the context of Parkinson’s disease biology. The authors identified a co-evolved LRRK2-regulatory module enriched for centrosome- and microtubule-associated cytoskeletal components and characterized new LRRK2 interactors. Defining an LRRK2–centrosome/microtubule network may clarify disease-relevant pathways and suggest targets for therapeutic modulation in familial and idiopathic PD.

Microglial Ccl4-Ccr5 signaling links systemic inflammation to synaptic loss and memory deficits in chronic kidney disease.
This animal and translational neuroscience study tested whether microglial Ccl4–Ccr5 signaling links systemic inflammation to synaptic loss and memory deficits in chronic kidney disease (CKD). Using a 5/6 nephrectomy rat model, the key finding was that CKD-associated cognitive impairment correlated with microglial activation and reduced synaptic density, with single-cell RNA sequencing and immunofluorescence supporting a mechanistic role for the Ccl4–Ccr5 axis in synaptic phagocytosis. Dental and Dentofacial Findings in a Child With CHOPS Syndrome Managed Under General Anesthesia.
This case report described dental and dentofacial findings in an 11-year-old male with CHOPS syndrome (a rare genetic disorder) managed under general anesthesia. FLNB and TTC26 regulate ciliary Hedgehog signaling to maintain intervertebral disc matrix homeostasis in adolescent idiopathic scoliosis.
This mouse genetics study examined whether gene-gene interactions between Filamin B (FLNB; Flnb) and tetratricopeptide repeat domain 26 (TTC26; Ttc26) regulate ciliary Hedgehog signaling to maintain intervertebral disc matrix homeostasis in adolescent idiopathic scoliosis (AIS). The key finding was that double heterozygous mice carrying heterozygous mutations in both Flnb and Ttc26 developed progressive spinal curvature during puberty, whereas single heterozygotes did not show the same phenotype. The scientific significance is that it implicates combined disruption of FLNB and TTC26 in ciliary Hedgehog pathway regulation as a mechanistic contributor to AIS-like spinal instability.

RNF19A::PLAG1-rearranged uterine mesenchymal tumor with myofibroblastic features and focal adipocytic differentiation: broadening the histologic and molecular genetic spectrum of PLAG1-rearranged uterine mesenchymal neoplasms.
The study investigated a rare RNF19A::PLAG1-rearranged uterine mesenchymal tumor with myofibroblastic features in a 50-year-old woman, using histopathology and immunohistochemistry to characterize the tumor. The tumor showed bland spindle-to-epithelioid cells in solid sheets/fascicles with fibrous stroma, low mitotic activity, no necrosis, and focal mature adipocytes, with co-expression of CD34, desmin, estrogen receptor, and progesterone receptor and retained RB1 expression. This expands the histologic and molecular spectrum of PLAG1-rearranged uterine mesenchymal neoplasms and supports RNF19A::PLAG1 as a diagnostic molecular driver even when adipocytic differentiation and myofibroblastic morphology are present.

Gene-specific long-term course, neurodevelopmental outcome and quality of life in patients with LIS1/PAFAH1B1-, DCX-, DYNC1H1-, TUBA1A- and TUBG1-related lissencephaly.

Oridonin as a novel KDM5C inhibitor alleviates clonal hematopoiesis-induced cardiac aging via H3K4me3-dependent SASP suppression.
The study investigated whether oridonin, a novel KDM5C inhibitor, can alleviate clonal hematopoiesis-induced cardiac aging in the context of TET2-driven CHIP and H3K4me3-dependent SASP regulation. Oridonin reversed CHIP- and aging-associated gene signatures and reduced cardiac aging phenotypes by suppressing SASP in a manner dependent on H3K4me3. These findings suggest KDM5C inhibition by oridonin may be a pharmacologic strategy to mitigate cardiovascular aging risk driven by TET2-CHIP.

Familial hypercholesterolaemia in children and adolescents: a European Atherosclerosis Society consensus statement.
This European Atherosclerosis Society consensus statement reviewed and standardized approaches for diagnosing and managing familial hypercholesterolaemia (FH) in children and adolescents, covering heterozygous FH (HeFH) and homozygous FH (HoFH). It emphasizes that early diagnosis and treatment in childhood can extend or normalize life expectancy and reduce premature ASCVD risk, particularly for HoFH where untreated risk is extreme even before adulthood. The guidance is significant for improving awareness, underdiagnosis, and undertreatment of pediatric FH across Europe.

Association of long-COVID with major adverse cardiovascular events and mortality: a real-world data cohort study.
Using a retrospective multicenter real-world cohort from the TriNetX electronic health record network, the study assessed whether long COVID (diagnosed 3–6 months after initial COVID-19) is associated with major adverse cardiovascular events (MACE) and all-cause mortality in adults aged ≥18 years. Long COVID was associated with increased risk of MACE and mortality compared with non–long COVID groups. Scientifically and clinically, it strengthens evidence that persistent post-acute sequelae of SARS-CoV-2 infection contribute to longer-term cardiovascular risk.

Time- and risk-dependent effects of dual antiplatelet therapy targeting the P2Y12 receptor after CABG: A network meta-analysis of graft permeability and clinical outcomes.
This network meta-analysis studied time- and risk-dependent effects of dual antiplatelet therapy (DAPT) targeting the P2Y12 receptor after coronary artery bypass grafting (CABG), comparing aspirin plus clopidogrel or ticagrelor versus aspirin-based regimens and rivaroxaban across follow-up periods and risk strata. Lipoprotein(a) and residual cardiovascular risks in Japanese patients with coronary artery disease who achieve guideline-recommended low-density lipoprotein cholesterol goals.
This multicentre retrospective observational study of 1581 Japanese patients with coronary artery disease (CAD) evaluated whether residual cardiovascular risk attributable to lipoprotein(a) [Lp(a)] persists in patients who achieve guideline-recommended low-density lipoprotein cholesterol (LDL-C) goals (<55 mg/dL). Major adverse cardiovascular events (MACE) were compared across Lp(a) strata (e.g., <30, ≥30 and <50, and ≥50 mg/dL) to determine risk differences despite LDL-C control. The study addresses how much Lp(a) contributes to residual risk when LDL-C is already well controlled, informing risk stratification in the absence of established Lp(a)-lowering therapies.

Association between METS-VF and hypertension in U.S.: a cross-sectional analysis of NHANES data.
This cross-sectional analysis of 13,822 U.S. participants from NHANES studied whether the Metabolic Score for Visceral Fat (METS-VF), calculated from age, sex, insulin resistance metabolic score, and waist-to-height ratio, is associated with hypertension. Higher METS-VF was associated with increased odds of hypertension after adjustment for confounders using logistic regression models. If validated longitudinally, METS-VF could provide a non-invasive clinical tool to identify individuals at higher cardiometabolic risk.

Cost-effectiveness of finerenone in heart failure with mildly reduced or preserved ejection fraction - Insights from FINEARTS-HF.
This analysis used FINEARTS-HF trial data (N=6,001) to evaluate the cost-effectiveness of finerenone in patients with heart failure with mildly reduced or preserved ejection fraction (HFmrEF/HFpEF) in the United States and Germany using a cohort-based Markov model. Finerenone was cost-effective under modeled horizons, with results sensitive to the analytic time horizon (in-trial vs lifetime). The findings support health-economic justification for finerenone adoption in HFmrEF/HFpEF populations beyond clinical efficacy alone.

The safety and efficacy of semaglutide in people with schizophrenia spectrum disorders: systematic review and meta-analysis of randomised controlled trials.
This systematic review and meta-analysis evaluated the safety and efficacy of the GLP-1 receptor agonist semaglutide (and related trials with tirzepatide) in people with schizophrenia spectrum disorders (SSDs) using placebo-controlled randomized controlled trials. Across included RCTs, semaglutide showed evidence relevant to weight and metabolic outcomes while assessing tolerability in SSD populations, addressing a gap in evidence versus the general population. Clinically, the results inform whether semaglutide can be used to reduce obesity/metabolic dysfunction–related morbidity in SSD patients.

Angiogenic T cells and cognitive function in older adults with type 2 diabetes treated with GLP-1 receptor agonists.
This cross-sectional study assessed whether circulating angiogenic T cells (“Tang cells”) relate to cognitive function in older adults with type 2 diabetes mellitus (n=154, ages 60–80) and whether associations differ by GLP-1 receptor agonist therapy. Tang cell levels were associated with cognitive performance, and GLP-1 receptor agonist treatment (GLP-1RA plus metformin vs metformin alone) appeared to influence this relationship (specific statistics truncated). If validated longitudinally, Tang cells could help link vascular/angiogenic biology to cognition and inform risk stratification in older T2DM patients on GLP-1RA.

Associations of coffee, alcohol, medication and supplement use with the metabolome and lipidome: an observational study of premenopausal women.
This observational study in 702 premenopausal women assessed how frequencies of coffee and alcohol intake, and use of medications and supplements, associate with the blood metabolome and lipidome using untargeted metabolomics and lipidomics. Beyond glycemic control: differential effects of empagliflozin and sitagliptin on insulin sensitivity and a shared increase in adropin in type 2 diabetes.

This preclinical mouse study tested whether colonization with soil-derived environmental (ENV) microbiota induces regulatory T (Treg) cells and protects against mouse colitis, metabolic disease, and sepsis. The key finding was that sustained colonization—especially with Gram-negative bacteria—shifted both local and systemic immunity toward anti-inflammatory responses characterized by increased Treg activity and improved disease outcomes. Association between C-reactive protein to lymphocyte ratio and depression: A population-based study.
This population-based study used NHANES 2015–2023 data (n=12,868) to examine whether the C-reactive protein to lymphocyte ratio (CLR; hs-CRP/lymphocyte count) is associated with depression and suicidal ideation measured by PHQ-9. Higher CLR was associated with greater depressive symptom burden and suicidal ideation after multivariable adjustment (details truncated in the abstract). If confirmed, CLR could serve as an accessible inflammatory biomarker to help stratify depression risk in the general population.

Emerging Corynebacterium Species Infections in the Shadow of Radiation Therapy: A Case Series in Cancer Patients.
This case series studied three culture-confirmed infections caused by Corynebacterium species in cancer patients receiving radiation therapy, including two Corynebacterium striatum skin/soft-tissue infections and one Corynebacterium matruchotii vaginal infection. Exploratory gut microbiota-host interaction in parkinson’s disease: integrative multi-omics analysis of metabolic and epigenetic pathways.

Microbiota-derived butyrate inhibits colonic epithelial pyroptosis and mitigates DSS-induced colitis via interacting with aryl hydrocarbon receptor.
This study tested whether microbiota-derived butyrate inhibits colonic epithelial pyroptosis and mitigates DSS-induced colitis through interaction with the aryl hydrocarbon receptor (AhR) in mice and cultured colonic epithelial cells (FHC) stimulated with LPS+ATP. The key finding was that butyrate reduced epithelial pyroptosis and colitis severity, and that this protective effect depended on AhR signaling (including evidence using GSDMD-knockout mice to implicate pyroptosis machinery). These results support butyrate/AhR as a mechanistic therapeutic axis for ulcerative colitis by targeting epithelial pyroptosis.

Structured Exercise Regimen in Pulmonary Hypertension-Right Ventricular Failure in an Ovine Model.
This JoVE study investigated the effects of a structured exercise regimen on pulmonary hypertension–right ventricular failure (PH-RVF) in an ovine model. Using eight Dorset cross sheep with PH-RVF induced by ligation of the left pulmonary artery and progressive occlusion of the main pulmonary artery with an inflatable cuff, the authors implemented an eight-week exercise protocol to characterize exercise-induced stress in this disease model. The model and regimen provide a controlled platform to study how exercise modulates PH-RVF pathophysiology and to inform future translational exercise strategies.

Determinants of tidal recruitment/derecruitment assessed by electrical impedance tomography in spontaneously breathing ARDS patients.

Impact of Biologics on Oscillometry Defined Small Airway Dysfunction in Uncontrolled Asthma.

[Use of inhaled nitric oxide during neonatal transport - a review of seven years of experience].

Genome-wide meta-analysis identifies genetic drivers of bile acid metabolism in intrahepatic cholestasis of pregnancy.
This genome-wide meta-analysis studied genetic drivers of bile acid metabolism in intrahepatic cholestasis of pregnancy (ICP) by combining genome-wide association study data from FinnGen, deCODE, the Estonian Biobank, the Danish Blood Donor Study, and Copenhagen Hospital Biobank (4,738 women with prior ICP and 436,834 female controls). The analysis identified 26 genome-wide significant loci (including 10 novel associations) and prioritized candidate genes using lead-SNP expression quantitative trait locus (eQTL) approaches (lead SNP expression quan[…]). These findings refine the genetic architecture of ICP and highlight bile-acid–related pathways that could inform risk stratification and mechanistic targets for therapy.

Antenatal N-acetylcysteine supplementation in pregnant women with impending preterm birth: a prospective randomized placebo-controlled trial.
This pilot prospective randomized placebo-controlled trial studied antenatal N-acetylcysteine (NAC) supplementation in pregnant women with impending preterm birth (singletons at moderate or severe risk; NCT03596125; n=20 NAC vs n=19 placebo). Mitochondrial Carrier SLC25A13 Drives Ferroptosis Resistance and Immune Evasion via a STAT3-IFI6 Circuit in Breast Cancer.
This study investigated how the mitochondrial carrier SLC25A13 influences ferroptosis and immune evasion in triple-negative breast cancer (TNBC). SLC25A13 was upregulated in breast cancer and associated with poor prognosis and reduced CD8+ T-cell infiltration; mechanistically, SLC25A13 promoted tumor growth and metastasis while suppressing ferroptosis and weakening CD8+ T-cell cytotoxicity via a STAT3–IFI6 circuit linked to enhanced complex I–dependent oxidative phosphorylation and reduced mitochondrial ROS. These results suggest targeting SLC25A13/STAT3–IFI6 signaling to restore ferroptosis and improve antitumor immunity in TNBC.

An in vivo parallelized reporter assay to uncover tissue-specific splicing regulatory sequences in a multicellular animal.
The study developed an in vivo parallelized reporter assay (PRA) in Caenorhabditis elegans neurons and muscle cells to identify tissue-specific intronic splicing enhancer and silencer sequences. Using thousands of randomized intronic sequences, the authors screened cis-regulatory motifs and identified nearly 200 sequences that regulate splicing in a tissue-dependent manner. Guidance for the assessment of the dietary intake and gastrointestinal or nutrition impact symptoms etiologic criterion of Global Leadership Initiative on Malnutrition: A modified-Delphi study.

Neoplasm Adverse Events Associated With Anti-Type 2 Biologics: An FAERS Database Pharmacovigilance Analysis Study.

The transmembrane segments of SARS-CoV-2 nsp3 govern viral intracellular trafficking.
This mechanistic virology study investigated how SARS-CoV-2 nonstructural protein 3 (nsp3) transmembrane segments control viral intracellular trafficking in the context of ER–ERGIC membrane targeting. Real-world pharmacovigilance analysis of drug-induced liver injury in 18-65 years: Based on the FDA adverse event reporting system (FAERS).
This real-world pharmacovigilance study analyzed drug-induced liver injury (DILI) epidemiology and drug-specific signals in adults aged 18–65 years using the FDA Adverse Event Reporting System (FAERS) from 2004–2024. The 2024 McDonald criteria for the diagnosis of multiple sclerosis: implications for clinical practice.

Real-World Safety and Retention of Tofacitinib in Elderly Versus Non-elderly Patients With Rheumatoid Arthritis: A Retrospective Cohort Study in Taiwan.

Age-dependent nuclear accumulation of a membrane-associated NAC transcription factor GER1 activates SA biosynthesis to promote leaf senescence in Arabidopsis.
This work investigated how the membrane-associated NAC transcription factor GERAS1 (GER1/GERAS1) controls age-dependent leaf senescence in Arabidopsis by regulating senescence-associated biosynthesis of salicylic acid (SA). GER1 expression increased with plant age and its protein translocated from the plasma membrane to the nucleus, with the activation-tagging mutant ger1-D causing early senescence and GER1 disruption delaying senescence. Plant viruses hijack and stabilize host Ubiquitin-Specific Protease 14 to prevent viral protein degradation for efficient infection.
This study examined how plant viruses prevent viral protein degradation by hijacking and stabilizing host Ubiquitin-Specific Protease 14 (UBP14) during infection in Nicotiana benthamiana. Ubiquitinome profiling showed reduced UBP14 ubiquitination and increased UBP14 accumulation across potyvirus infections, and the potyviral cylindrical inclusion (CI) protein bound UBP14 to suppress its ubiquitination, recruit it to viral replication complexes, and deubiquitinate/stabilize viral replication components. These results reveal a conserved viral strategy to enhance infection efficiency by directly manipulating host deubiquitination machinery.
