Apolipoprotein B-containing lipoproteins: distinct roles in atherosclerosis revealed by plasma proteomic profiling.

Perinatal lead (Pb) exposure and alterations to amyloid beta and metabolic profiles in the brain.

Longitudinal plasma phosphorylated tau217 classification and the association with Alzheimer’s disease progression.

This multi-cohort study evaluated the longitudinal diagnostic and prognostic performance of baseline plasma phosphorylated tau217 (p-tau217) for predicting amyloid-β PET (Aβ-PET) positivity and subsequent Alzheimer’s disease progression in 2117 individuals. Group A Streptococcus molecular point of care testing in a paediatric emergency department.

DLST Succinylation-Mediated Mitochondrial Metabolic Remodeling and Cuproptosis Resistance Promote Malignant Progression of Lung Adenocarcinoma.

Space Jam-Ming: Generating Interstitial Space Using Fragmented Granular GelMA to Investigate Novel Paradigms of Cancer Metastasis.

This study developed “Space Jam-Ming,” a fragmented granular GelMA hydrogel platform to create interstitial space with controlled mechanical properties for modeling cancer metastasis. Extracellular matrix remodeling upregulates hippocampal neurogenic niche stiffness and impairs neurogenesis in Alzheimer’s disease.

This study examined whether extracellular matrix (ECM) remodeling increases hippocampal neurogenic niche stiffness and impairs neurogenesis in Alzheimer’s disease using 5×FAD mice and human AD samples. ANGII Initiated HSF2 Trans-Activating HIF-1α Through Induction of ER Stress to Promote Cardiac Hypertrophy.

This study tested whether angiotensin II (ANGII) promotes cardiac hypertrophy by inducing ER stress that enables HSF2 to trans-activate HIF-1α in cardiomyocytes, using both in vitro and in vivo models. ANGII increased HIF-1α expression via an HSF2-dependent transcriptional mechanism associated with ER stress, supported by RT-PCR, Western blotting, immunocytochemistry/immunohistochemistry, luciferase reporter assays, and chromatin immunoprecipitation (ChIP). Natural Product Toosendanin Suppresses the Malignant Development of Skin Melanoma by Targeting BNC2 for Degradation.

This study tested the natural product toosendanin (TSN) in skin melanoma models and investigated its molecular target. TSN inhibited melanoma proliferation and cell-cycle progression, induced apoptosis, and suppressed metastasis, and it was shown to target basonuclin 2 (BNC2) for degradation, thereby suppressing PIK3CA transcription. This cross-sectional study evaluated whether waist circumference outperforms body mass index (BMI) for cardiometabolic risk discrimination and reclassification in 572 consecutive patients with psoriasis and/or psoriatic arthritis. A case of diffuse large B-cell lymphoma achieving complete remission with epcoritamab after pseudoprogression.

[Thiotepa-busulfan-fludarabine conditioning in allogeneic hematopoietic stem cell transplantation for blast-phase myeloproliferative neoplasm].

This Lancet Series reviewed how adverse pregnancy outcomes (APOs)—including hypertensive disorders of pregnancy, gestational diabetes, and preterm birth (<37 weeks)—identify women with substantially higher long-term cardiovascular morbidity and mortality risk. It proposes that excess risk reflects both pre-existing cardiometabolic/genetic susceptibility and the haemodynamic/metabolic stressors of pregnancy that accelerate future disease. Scientifically and clinically, the synthesis supports using APO history for cardiovascular risk stratification and long-term surveillance after pregnancy.

Lipoprotein(a) and Atherosclerotic Cardiovascular Disease Among Adults Aged 20 to 100 Years.

This study evaluated whether the association between high lipoprotein(a) [Lp(a)] and atherosclerotic cardiovascular disease (ASCVD) differs by age and sex in 103,341 adults (Copenhagen General Population Study) aged 20–100 years, including 18,322 participants aged 70–100 years. Single-cell and spatial transcriptomic profiling identifies a fibroblast-like endothelial state and the MDK-NCL signaling axis in prostate cancer microenvironment remodeling.

This translational oncology study used single-cell RNA sequencing and spatial transcriptomics (integrated with bulk transcriptomic cohorts) to characterize endothelial heterogeneity in the prostate cancer microenvironment and identify a fibroblast-like endothelial (FLE) state. National survey of patients with neuromuscular disease and COVID-19 in Japan.

Brugada Syndrome in the Pediatric Population: Diagnostic Challenges and Clinical Outcome-Results of the German Multicenter Study COGIA.

Low-Voltage Ablation in Persistent Atrial Fibrillation: The IDEAL-AF Randomized Clinical Trial.

Autophagic degradation of RAB8A promotes ferroptosis through dysregulation of TFRC-mediated iron uptake.

This study investigated how selective autophagy regulates ferroptosis by identifying RAB8A as an ATG5/ATG7-dependent autophagic substrate in cancer cells under ferroptotic stress. Ferroptotic stimuli induced RNF126-dependent polyubiquitination of RAB8A followed by SQSTM1-mediated autophagic degradation, and loss of RAB8A sensitized cells to ferroptosis while a degradation-resistant mutant (RAB8AQ67L) suppressed this effect. Efficacy and safety of upadacitinib for Crohn’s disease in patients in East Asia: a post hoc analysis of randomized phase 3 studies.

A post hoc analysis of two randomized phase 3 induction trials (U-EXCEL and U-EXCEED) and the 52-week maintenance trial (U-ENDURE) assessed the efficacy and safety of the oral JAK inhibitor upadacitinib in adults with moderate-to-severe Crohn’s disease in East Asia. The analysis evaluated outcomes after induction with upadacitinib 45 mg versus placebo and maintenance with upadacitinib 15 mg or 30 mg versus placebo among responders. Early SOX9 Activation Primes Hippo-YAP/TAZ Rewiring During Glioblastoma Stemness Acquisition.

Identifying Patients With Prostate Cancer Who Benefit Most From Routine Cardiovascular Specialist Referral.

This analysis studied the RADICAL PC-2 pragmatic randomized controlled trial population of patients with prostate cancer or receiving androgen-deprivation therapy to identify subgroups most likely to benefit from routine cardiovascular specialist referral. COVID-19 Vaccination and Risk of Post-COVID-19 Cardiovascular Disease: A Population-Based Cohort and Target Trial Emulation Study.

This population-based cohort study and target trial emulation evaluated whether COVID-19 vaccination, including timing relative to SARS-CoV-2 infection, is associated with reduced long-term post-COVID cardiovascular disease risk in 2,391,456 adults in Victoria, Australia (2019–2025). SerpinA3 is an Endogenous TGF-β Receptor Antagonist that Attenuates Cardiac Fibroblast Activation and Fibrotic Remodeling.

Time in blood pressure range and cardiovascular outcomes in HFmrEF/HFpEF: a pooled participant-level analysis of four large-scale trials.

Gene Silencer Therapy in Transthyretin Amyloid Cardiomyopathy: A Meta-Analysis of Outcomes Trials.

This meta-analysis synthesized outcomes from randomized, placebo-controlled phase 3 trials of transthyretin (TTR) gene-silencing therapies in patients with transthyretin amyloid cardiomyopathy (ATTR-CM), with attention to baseline use of TTR stabilizers. SIRT1 Silences L1 Retrotransposons by Stabilizing Heterochromatin-Modifying Complexes.

This study examined how the sirtuin deacetylase SIRT1 regulates LINE-1 (L1) retrotransposition during cellular senescence, focusing on L1 activity and heterochromatin-modifying complexes. Cytokine-associated ROS and Ca2+ dysregulation in severe inflammatory cardiomyopathy.

This study investigated cytokine-associated mechanisms of reactive oxygen species (ROS) generation and Ca2+ dysregulation in severe inflammatory cardiomyopathy (iCMP) using cytokine profiling in patients with biopsy-proven iCMP (n=63) and validation in a national cohort (n=425), followed by induced human cardiomyocyte experiments. It identified key cytokines enriched in severe iCMP and showed that these cytokines promote cardiomyocyte injury by driving ROS production and disrupting calcium homeostasis. Mechanistically, the work highlights cytokine–ROS–Ca2+ signaling as a potential therapeutic target in severe iCMP.

In the RECOVER-VITAL phase 2 randomized, double-blind, placebo-controlled trial, adults with long COVID symptoms lasting ≥12 weeks across cognitive, autonomic, or exercise phenotypes were treated at 69 US sites with the SARS-CoV-2 antiviral nirmatrelvir-ritonavir versus placebo. The study tested whether nirmatrelvir-ritonavir improves long COVID symptoms by targeting a viral persistence hypothesis. If effective, this would support an antiviral strategy for persistent post-acute sequelae of SARS-CoV-2 infection in a phenotype-defined adult population.

Refractory Azole-Resistant Candida albicans Renal and Perirenal Abscess Requiring Nephrectomy.

Military Service and Survival Among Persons With ALS in the U.S. National ALS Registry, 2011-2023.

Antidepressant effects of facial botulinum toxin a injections: Insights from randomized controlled trials.

This systematic review and meta-analysis evaluated whether facial injections of botulinum toxin A (BoNT/A) reduce depressive symptoms compared with placebo and the active antidepressant sertraline in randomized controlled trials. A single-cell meta-analysis evidences transposable element dysregulation in sex-based differences in Parkinson’s disease.

Intrathecal semaglutide attenuates burn injury-induced pain in mice through a spinal GLP-1R-linked enkephalin/δ-opioid receptor pathway.

This mouse study tested whether intrathecal semaglutide attenuates burn injury-induced pain (BIP) via spinal glucagon-like peptide-1 receptor (GLP-1R) signaling coupled to an enkephalin/δ-opioid receptor pathway. Interval-Based Aerobic Exercise and Behavioral Intervention for Sleep Disturbances in Children With Autism Spectrum Disorder: A Randomized Controlled Trial.

This randomized controlled trial studied whether interval-based aerobic exercise plus parent behavioral education (IAE-PBE) improves sleep and related outcomes in 28 children with autism spectrum disorder (ASD) and sleep disturbances, compared with parent behavioral education (PBE) alone. YTHDC1 Orchestrates Glucose and Glutamate Rewiring to Overcome Lethal Metabolic Stress in Triple-Negative Breast Cancer.

This study examined the role of the m6A reader YTHDC1 in triple-negative breast cancer (TNBC) metabolic adaptation under lethal metabolic stress. YTHDC1 was specifically overexpressed in TNBC and orchestrated m6A-dependent rewiring by stabilizing GLUT3 mRNA to sustain glucose uptake/NADPH production while reducing ATF4 mRNA stability to suppress SLC7A11-mediated cystine uptake. Targeting the YTHDC1–m6A–GLUT3/ATF4 axis may represent a therapeutic approach to overcome metabolic stress in TNBC.

Charting Endocrine Progenitors Across Species and Organs.

This cross-species, cross-organ study mapped endocrine progenitors (EPs) across mouse and human pancreas and intestine using single-cell transcriptomics and epigenomics, complemented by mouse bulk proteomics. Deubiquitination of Vangl by USP6 and USP32 Regulates Planar Cell Polarity Signaling.

AARS1-Mediated H3K27 Lactylation Rewires Glycolysis to Sustain Aggressive and Recurrent Bladder Cancer.

This study investigated how alanyl tRNA synthetase 1 (AARS1) drives aggressive and recurrent bladder cancer by rewiring glycolysis through H3K27 lactylation. AARS1 was upregulated in bladder cancer, enriched in muscle-invasive and recurrent tumors, and promoted proliferation, EMT, invasion, apoptosis resistance, tumor growth, and lung colonization while enhancing PI3K pathway output, glycolytic flux, and lactate production. These findings position the AARS1–H3K27 lactylation–glycolysis axis as a mechanistic driver and potential therapeutic target for recurrence-prone bladder cancer.

IFI27 and BAX are Essential for GSDME-Mediated Myeloma Cell Pyroptosis.

This mechanistic study examined how IFI27 and BAX regulate GSDME-mediated pyroptosis in multiple myeloma (MM) cells. It found that the interferon-alpha–inducible protein IFI27 is upregulated in pyroptotic MM cells and that IFI27 overexpression triggers pyroptosis in MM cell lines and newly diagnosed MM cells in a GSDME-dependent manner, requiring IFI27-driven mitochondrial disruption and the pro-apoptotic factor BAX. The work identifies IFI27 and BAX as essential components of a potential pyroptosis-based therapeutic pathway for MM.

WTAP Transcriptional Suppression by KLF9 Drives Osteoclastogenesis via M6A-Mediated Regulation of CSF1R Signaling in Estrogen-Deficient Osteoporosis.

This study investigated how WTAP transcriptional suppression by KLF9 drives osteoclastogenesis in estrogen-deficient osteoporosis via m6A-dependent regulation of CSF1R signaling. In mice, myeloid-specific Wtap knockout worsened osteoclast formation and bone loss, and mechanistically WTAP promoted m6A deposition on Csflr mRNA to enable YTHDF2-mediated degradation and downregulate CSF1R expression, while KLF9 induction suppressed WTAP. The findings link KLF9–WTAP–m6A–YTHDF2 control of CSF1R to osteoclast activation, suggesting epitranscriptomic targets for osteoporosis therapy.

NAD+ precursor treatment prevents cardiomyopathy but disrupts erythroid maturation in mitochondrial progeria.

This preclinical study tested whether nicotinamide riboside (NR), an NAD+ precursor, prevents cardiomyopathy in “mutator” mice with mitochondrial progeria while assessing tissue-specific effects on erythropoiesis. NR treatment prevented or reduced cardiomyopathy but disrupted erythroid maturation, worsening anemia, and in bone marrow it caused reductive stress with altered NADH/NADPH and impaired heme biosynthesis. The results indicate that NAD+ restoration can have beneficial cardiac effects yet harmful hematopoietic consequences, underscoring the need for careful tissue-targeted strategies.

Phase 3 Randomized Trial Results of DTX401 AAV Gene Therapy for the Treatment of GSDIa.

This phase 3, double-blind, randomized, placebo-controlled trial evaluated DTX401, an AAV8 vector delivering the human G6PC gene, in patients aged ≥8 years with glycogen storage disease type Ia (GSDIa) due to biallelic pathogenic G6PC variants. The primary efficacy endpoint compared percent change from baseline to week 48 in daily cornstarch intake between DTX401 and placebo groups (with a subsequent crossover after the primary efficacy analysis period). If positive, DTX401 would represent a gene-therapy approach targeting the underlying G6PC deficiency to improve metabolic control in a rare, life-threatening pediatric/adolescent/adult population.

Treatment of multiple sclerosis with peptide-coupled red blood cells induces antigen-specific T regulatory cells.

This PNAS open-label, dose-escalation phase Ib clinical trial tested antigen-specific immune tolerance in multiple sclerosis by administering autologous red blood cells coupled to seven immunodominant myelin peptides (pcRBCs). The key findings were that pcRBCs were safe and well tolerated, several clinical/imaging measures remained stable with reduced neurofilament light chain suggesting decreased brain inflammation, and single-cell RNA sequencing showed expansion of antigen-specific T regulatory cells. Scientifically and clinically, it supports a peptide-coupled RBC strategy to induce antigen-specific Tregs as a potential organ-targeted immunotherapy for MS.

CAR-Engineered Cell Therapies Beyond Cancer: Reprogramming Fibrosis and Immune-Mediated Inflammation.

Biomaterials in Retinal Surgery: Engineering the Future.

This narrative expert review summarized current and emerging biomaterials used in retinal surgery, including vitreous tamponade agents, retinal prostheses, retinal pigment epithelium (RPE) transplant scaffolds, intraocular drug delivery platforms, and gene therapy vectors, with emphasis on oxime-crosslinked hyaluronic acid vitreous substitutes. An Immune-Activated Co-culture Model of Patient-Derived Gastric Cancer Organoids and Peripheral Blood Mononuclear Cells for Personalized Chemotherapy Screening.

This study developed an immune-activated co-culture model using patient-derived gastric cancer organoids and autologous peripheral blood mononuclear cells (PBMCs) to enable personalized chemotherapy screening after immunotherapy. Immune activation was achieved using anti-PD-1-loaded gold nanocages (aPD-1@Au NCs), and the model was designed to preserve immune–tumor interactions while testing subsequent chemotherapy responses. Real-world adherence and healthcare resource utilization following transition to aripiprazole once-every-2-months from oral or once-monthly aripiprazole among patients diagnosed with schizophrenia in the United States.

This retrospective real-world claims study evaluated adherence and healthcare resource utilization in U.S. adults with schizophrenia who transitioned from oral aripiprazole or once-monthly aripiprazole monohydrate to aripiprazole monohydrate 2-month ready-to-use (Ari 2MRTU). Following the transition, adherence improved (PDC and MPR increased) and healthcare resource utilization outcomes were assessed in the 6 months before versus after index. The findings suggest that extending dosing to every 2 months may improve medication-taking behavior and potentially reduce downstream utilization burden in schizophrenia care.

Hypereosinophilic syndrome and suspected myeloid neoplasia in relapsed Hodgkin lymphoma during immune checkpoint inhibitor therapy: a rare case.

This case report studied immune-related adverse events in a 56-year-old man with stage IV nodular sclerosis classic Hodgkin lymphoma who relapsed after multiple prior therapies and then received pembrolizumab plus gemcitabine/vinorelbine/liposomal doxorubicin (GVD). After achieving a complete metabolic response, he developed marked leukocytosis with profound eosinophilia consistent with hypereosinophilic syndrome and suspected myeloid neoplasia during immune checkpoint inhibitor therapy. The report highlights a rare but clinically important spectrum of hematologic toxicity that can mimic or reveal underlying myeloid disease during PD-1 blockade.

Curcumin Suppresses Growth of Recurrent Colorectal Adenoma After Endoscopic Resection: A Randomized, Double-Blind, Placebo-Controlled, Multicenter Trial.
